Dana-Farber Cancer Institute Research Technician-Gu Lab in Boston, Massachusetts

Dr. Xin Gu’s laboratory at the Dana-Farber Cancer Institute is seeking a motivated research technician, who is interested in learning how to do mechanistic molecular and cellular biology research as well as contributing to exciting ongoing projects. Proteins typically require ubiquitin conjugation for degradation by a macromolecular protease complex called the proteasome. The Gu lab aims to elucidate the molecular mechanism of the ubiquitin-independent proteasomal degradation and understand its physiological significance. Despite decades of sporadic reports on individual proteins being degraded by the proteasome independently of ubiquitin conjugation, no general mechanism was described. Our recent discovery has elucidated a novel pathway, termed the midnolin-proteasome pathway (MPP), that can degrade hundreds of nuclear proteins key for transcriptional regulation, bypassing the ubiquitin requirement (https://www.science.org/doi/10.1126/science.adh5021 ). This finding opened many exciting areas of ongoing exploration. These include characterizing the MPP using cells and purified proteins as well as determining the purpose of the MPP for organismal physiology. We will also establish tools for hijacking the MPP for therapeutic potential.

The ideal individual will learn how to conduct scientific research from experimental design to practical execution. Enthusiasm towards science, collaborative nature, and interpersonal skills are essential. We seek diverse skills including molecular cloning, mammalian cell culture, mouse neuron dissection, chromatin immunoprecipitation (ChIP), protein-protein interaction detection, proteomics, western blotting, and flow cytometry. This position is a great steppingstone for individuals who aim to pursue biomedical research.

  • Learning and conducting molecular and cellular assays, such as cloning, cell culture (including neurons), various immunoprecipitation techniques, various sequencing techniques, proteomics, western blotting, and flow cytometry.

  • Keeping detailed experimental notes and organizing obtained data.

  • Assisting with experimental design and reagent preparation.

  • Assisting with data analysis and interpretation.

  • Presenting experimental results in a weekly meeting within the laboratory to obtain feedback and make further plans.

  • Attending seminars for learning purposes within the Harvard Medical School research environment.

  • Must be passionate and fearless about learning and performing biological research.

  • Bachelor's Degree required (or expected to be obtained prior to start date).

  • Prior molecular and cellular research experience, or mouse experience is preferred.

  • Must have excellent communication skills (written and verbal) and be able to work with a wide variety of faculty and staff.

  • A minimum time commitment of 2 years is highly preferred.
