CRISPR Therapeutics Senior Bioanalytical Scientist, Non-viral Delivery in Cambridge, Massachusetts

Company Overview

At CRISPR Therapeutics, we are focused on developing transformative gene-based medicines for serious human diseases. We are rapidly translating our CRISPR/Cas9 gene editing platform into therapies to treat hemoglobinopathies, cancer, diabetes, and other diseases. Our multi-disciplinary team of world-class researchers and drug developers is committed to translating CRISPR/Cas9 technology into breakthrough human therapeutics. Our lead program targeting the blood diseases β-thalassemia and sickle cell disease has entered clinical testing, and our immuno-oncology programs focused on CRISPR/Cas9-edited allogeneic CAR-T cell therapies have the potential to deliver the next generation of cancer therapies to patients.

Job Summary

We are seeking an experienced Research Scientist to expand our capabilities in analytical biochemistry. The successful candidate will leverage their technical expertise in analytical chromatography and mass spectrometry to advance our gene editing platform by providing key insights into the synthesis, purification, and structure-activity relationships of oligonucleotides, including DNA and RNA.

  • Develop and expand internal capabilities for the characterization of nucleic acid therapies and components thereof (DNA, gRNA, and mRNA) with an emphasis on chromatographic methods (IEX, IPRP HPLC/UPLC & SEC), mass spectrometric methods (LC-MS, LC-MS/MS), and electrophoretic methods (agarose, PAGE, CE)

  • Develop methods for the extraction and quantitation of nucleic acid and their metabolites from cells and complex biologic matrices (serum, plasma, and tissues) using LC-MS

  • Develop enzymatic and cell-based assays for the characterization of therapeutic nucleic acids

  • Apply knowledge of biochemistry and molecular biology techniques for the physical and enzymatic modification of therapeutic nucleic acids

  • Perform validation, QC, and optimization of new nucleic acid reagents and precursors

  • Ensure proper operation and maintenance of laboratory instruments, including HPLC/UPLC systems and mass spectrometers

  • Maintain an accurate and up-to-date record of experiments and results in the form of ELN entries, reagent inventory and registry, and written reports

  • Communicate key findings in group and cross-functional team meetings

  • Contribute to regulatory and IP filings

  • Perform laboratory work in safe, efficient manner to ensure a safe working environment for all colleagues

Minimum Qualifications

  • PhD in chemistry, biochemistry or related field, and 5+ years of highly relevant laboratory experience

  • Deep understanding of analytical methods required to support development of novel nucleic acid chemistries and delivery modalities

  • Extensive hands-on experience with the following:

  • Analyte extraction from biological matrices

  • An educator’s mindset with an eagerness to invest in communication with scientists and senior leaders, and a passion for mentorship of junior associates

  • A learner’s mindset with a commitment to personal growth and a drive to broaden technical skills and scientific knowledge

  • The ability to think strategically, efficiently manage time and workflows, and work both independently and collaboratively in a cross-functional team environment

Preferred Qualifications

  • Experience managing and mentoring junior research associates

  • Proficiency with the extraction and quantification of lipids and their metabolites from cells and complex biologic matrices (serum, plasma, tissue) using LC-MS
